چکیده

In order to study the hydrolysis, enzymatic or chemical, of the plasma and urinary steroid glucuronides, as well as the biosynthesis, the isolation of the individual 17-oxosteroids is an absolute necessity. Schneider and Lewbart (1959) employed counter-current distribution techniques to isolate several steroid glucuronides. Androsterone glucuronide was isolated by Edwards and Kellie (1956) and by Crepy, Jayle, and Meslin (1957); and the aetiorcholanolone salt was isolated by Baulieu (1960). Hadd and Dorfman (1963) isolated androsterone glucuronide from a patient producing a massive amount of it. Since then both 5 ca (Staib and Doenges, 1960) and 5,B (Staib and Doenges, 1960; Hadd, 1958) forms of the 17-oxosteroid glucuronide have been synthesized. The object of the present work was to develop a method of separating the major 17-oxosteroid glucuronides on a large scale and economically. In the techniques developed inexpensive and easily available apparatus is used.

The indirect analysis of 17-oxosteroid glucosiduronates and sulphates in human urine.

1. An analytical procedure has been developed for the determination of urinary 17-oxosteroid sulphates and 11-deoxy-17-oxosteroid glucosiduronates. 2. By submitting urine to the following operations: (a) extraction of the 17-oxosteroid conjugates into chloroform (in the presence of pyridinium sulphate), (b) solvolysis (in the absence of acid) of the steroid sulphates, (c) separation of the ster...

Effect of SodiumSulfateon the Hydrolysisof 17-Hydroxycorticosteroid- andp-Nitrophenyl-glucuronides with fl-GlucuronidasePreparationsfrom BovineLiver

Previous papers dealt with the discovery that hydrolysis of 17-hydroxycorticosteroid glucuronides in urine with fi-glucuronidase preparations from bovine liver is increased by adding sodium sulfate to the incubation medium. Here, we conclude that sodium sulfate not only increases the activity of bovine liver /9-glucuronidase on 17-hydroxycorticosteroid glucuronides and p-nitrophenyl glucuronide...

The analysis of urinary 17-oxo steroids by gradient elution.

The present work was initiated by the observation of Reifenstein, Homburger & Dobriner (1950) that 3a: 1 1l-dihydroxy-5p-androstan-17-one (1 l,B-hydroxyaetiocholanolone), a minor component of the urinary 17-oxo steroids, was significantly associated with neoplastic growth. The re-examination of this problem has led to the development of a micro method for the quantitative separation and determi...

Direct determination of four sulfates and seven glucuronides of 17-oxosteroids in urine by fluorescence "high-performance" liquid chromatography.

We describe a direct method for determining four sulfates and seven glucuronides of 17-oxosteroids (17OS) in urine without hydrolysis, by use of "high-performance" liquid chromatography (HPLC) with fluorometric detection. After pretreatment of urine samples with a Sep-Pak C18 cartridge, four 17OS sulfates and seven 17OS glucuronides in the pretreated urine samples were reacted with tetrapentyla...

Simple, Fast and Reliable Liquid Chromatographic and Spectrophotometric Methods for the Determination of Theophylline in Urine, Saliva and Plasma Samples

In this study, a high-performance liquid chromatographic method (HPLC) and UV spectrophotometric method were developed, validated and applied for the determination of theophylline in biological fluids. Liquid- liquid extraction is performed for isolation of the drug and elimination of plasma and saliva interferences. Urine samples were applied without any extraction. The chromatographic separat...
